2、rvisor:Prof. SUN Zhen- zhu Abstract Objective: To analyze the histology features of endometrial lesion precancerous and endometrioid adenocarcinoma(EC), and explore the expression of PTEN, Her- 2 and

3、 Glut- 1 in endometrioid adenocarcinoma and their role in tumorigenesis. Methods: Envision immunohistochemistry was used to detect the protein expressions of PTEN, Her- 2 and Glut- 1 in 144 cases of e

4、ndometrial lesions.All cases were reviewed and a total of 84 cases of endometrial hyperplasia were selected and reclassified according to EIN diagnostic criteria. Results: (1) 50 EIN lesions and

5、 34 benign hyperplasia cases were tested among 84 previously diagnosed endometrial hyperplasia.(2) The negative expression rate of PTEN in benign hyperplasia, EIN and endometrial carcinoma were 35.3%, 64

6、.0% and 66.7% respectively; There were not significant differences among EIN and endometrioid adenocarcinoma(P>0.05),But benign hyperplasia and EIN, or benign hyperplasia and endometrioid adenocarc

7、inoma had significant differences(P<0.05).(3) The positive expression rate of Her- 2 and Glut- 1 in benign hyperplasia, EIN and endometrial carcinoma were 0%, 26.0%, 61.7% and 8.8%, 80%, 100%. Her-

8、 2 and Glut- 1 have significant differences among benign hyperplasia, EIN and endometrioid adenocarcinoma(P<0.05). (4) The expression of Her- 2 was correlected with muscles invasion, nationality and vessel

9、invasion(P<0.05). The expression of Glut- 1 had significant differences in histological grade, clinical stage,tumor’ size and vessel invasion(P<0.05). Conclusions: The diagnostic criteria of EIN was emphasi

10、zed that lesions size, histology structure and cytology feature were more important for the diagnosis, with good reproducibility and high consistency; PTEN,Her- 2 and Glut- 1 may play a significance role in th

11、e development of endometrioid adenocarcinoma; The combined use of PTEN, Her- 2, Glut- 1 and the histology features of diagnostic criteria is valuable in distinguishing endometrioid adenocarcinoma from endom

12、etrial lesion precancerous with; The positive expression of Her- 2 and Glut- 1 were correlected with the poor Prognosis of endometrioid adenocarcinoma. Key words: PTEN; Her- 2; Glut- 1; endometrial;E
