簡(jiǎn)介:ATHESISDISSERTATIONSUBMITTEDTOZHENGZHOUUNIVERSITYFORTHEDEGREEOFMASTERPROMOTERS,CLUSTERINGCHARACTERISTICSANDEVOLUTIONARYHISTORYANALYSISINMAIZEBYPENGYANGZHAOSUPERVISORPROFZENGYUANTIANBIOTECHNOLOGYCOLLEGEOFLIFESCIENCEMARCH2014摘要摘要MICRORNA簡(jiǎn)稱MIRNA,是一類在進(jìn)化中高度保守的小分子非編碼RNA,在真核生物的基因表達(dá)調(diào)控方面發(fā)揮重要作用。MIRNA的成熟序列與AGO蛋白有選擇性結(jié)合形成RISC復(fù)合體RNAINDUCEDSILENCINGCOMPLEX,與相應(yīng)靶基因的MRNA結(jié)合使其表達(dá)受到抑制或降解。MIRNA優(yōu)先結(jié)合的靶標(biāo)涉及的調(diào)節(jié)功能廣泛,集中于生長(zhǎng)發(fā)育、信號(hào)轉(zhuǎn)導(dǎo)以及脅迫相應(yīng)。本文研究了玉米B73REFGENV2參考染色體組中161個(gè)MIRNA的164個(gè)MIRNA基因位點(diǎn),其中最大的一個(gè)家族是MIRL69共有17個(gè)成員,MIRL62、MIR408、M1R482只被檢測(cè)到1個(gè)成員存在于該染色體中,為成員數(shù)最少的家族。真核生物細(xì)胞中沒(méi)有操縱子,在基因表達(dá)的調(diào)控過(guò)程中順式作用元件結(jié)合相應(yīng)的反式作用因子對(duì)RNA聚合酶的轉(zhuǎn)錄具有調(diào)控作用,這些CARECISACTINGREGULATORYELEMENT在基因序列側(cè)面存在與相應(yīng)的基因往往具有一定的距離,能夠與特定的反式作用因子相互作用調(diào)控真核生物基因表達(dá)的時(shí)間與空間特異性?;蚍执噩F(xiàn)象是基因在染色體上的一種存在形式,位于同一個(gè)簇的基因常常是由兩個(gè)或者兩個(gè)以上的家族成員組成,分布在同一個(gè)簇中的基因往往具有協(xié)同調(diào)控作用。全文有以下幾個(gè)重要的結(jié)論L、通過(guò)TSSP預(yù)測(cè)得到的相應(yīng)的啟動(dòng)子,根據(jù)這些TSS位點(diǎn)上游20KB域內(nèi)啟動(dòng)子的數(shù)量將玉米基因組REFGENV2中的161個(gè)MIRNA的164個(gè)基因分為四組,分別為未檢測(cè)到啟動(dòng)子,僅含有一個(gè)啟動(dòng)子,含有兩個(gè)啟動(dòng)子的和含有三個(gè)啟動(dòng)子或者更多。玉米MIRNA順式調(diào)控元件在MIRNA基因TSS上游10KB的區(qū)域內(nèi)容納的轉(zhuǎn)錄因子結(jié)合位點(diǎn)的數(shù)量平均為220左右。部分順式調(diào)控元件在所有的MIRNA基因上游區(qū)域廣泛存在,其中排名前十的是CACTFTPPCAL、CURECORECR、DOFCOREZM、ACGTATERDL、GTGANTGLO、EBOXBNNAPA、MYCCONSENSUSAT、GATABOX、ARRLAT、CAATBOXL,與脅迫相關(guān)的調(diào)控元件有CURECORECR涉及銅離子脅迫和氧脅迫、ACGTANRDL涉及早期脫水脅迫、MYCCONSENSUSAT涉及冷脅迫、ARRLAT涉及脅迫與非共生血紅蛋白相關(guān)。CACTFTPPCAL的數(shù)量最多與磷酸烯醇丙酮酸羧化酶相關(guān),在C4植物固定C02的過(guò)程中發(fā)揮重要作用。DOFCOREZM與DOF蛋白相關(guān),在植物的生長(zhǎng)發(fā)育過(guò)程中發(fā)揮重要作用。
