簡(jiǎn)介:華中農(nóng)業(yè)大學(xué)碩士學(xué)位論文豬傳染性胃腸炎病毒S基因表達(dá)的研究及其原核表達(dá)產(chǎn)物的生物學(xué)活性分析姓名楊樂(lè)樂(lè)申請(qǐng)學(xué)位級(jí)別碩士專(zhuān)業(yè)預(yù)防獸醫(yī)學(xué)指導(dǎo)教師吳斌郭福生200505012002級(jí)碩士研究生學(xué)位論夕良好的反應(yīng)原性。以表達(dá)的融合蛋白為抗原初步建立了間接酶聯(lián)免疫吸附試驗(yàn)EUSA方法;用方陣滴定確定其最佳抗原包被濃度分別是5/ZG/ML、1嘰G/ML,最佳血清稀釋倍數(shù)均為140。為了更好地消除非特異性反應(yīng),增強(qiáng)表達(dá)產(chǎn)物的抗原性,利用桿狀病毒表達(dá)系統(tǒng)可進(jìn)行翻譯后修飾等特點(diǎn),我們?cè)噲D在SF9昆蟲(chóng)細(xì)胞中進(jìn)行目的基因的表達(dá)。目前已成功構(gòu)建了重組桿狀病毒穿梭表達(dá)載體BACMID.S1、BACMIDS1.2,為目的基因桿狀病毒表達(dá)的進(jìn)一步研究奠定了基礎(chǔ)。本研究在TGEV病毒增殖的基礎(chǔ)上,克隆了TGEVS基因的兩個(gè)不同區(qū)段,構(gòu)建了原核表達(dá)載體并進(jìn)行了表達(dá),利用原核表達(dá)產(chǎn)物柙步建立了TGEV和PRCV的間接ELISA鑒別診斷方法。同時(shí)成功構(gòu)建了菱組桿狀病毒穿梭表達(dá)載體。這些工作為進(jìn)一步研究TGEV和PRCV的流行和變異、開(kāi)發(fā)新的診斷方法奠定了基礎(chǔ),同時(shí)為其它冠狀病毒的研究提供了參考。關(guān)鍵詞TGEV{PRCV;S基因;原核表達(dá);BAC.TO.BAC鑒別診斷

簡(jiǎn)介:丁香因枝葉繁茂、花色淡雅而清香、耐寒、耐旱及土壤要求不高,因此是世界重要綠化、美化、香花樹(shù)種。丁香品種繁多,一些品種之間性狀比較相近,容易混淆。通過(guò)對(duì)包頭地區(qū)栽培的十四個(gè)品種進(jìn)行生長(zhǎng)發(fā)育習(xí)性指標(biāo)的調(diào)查和統(tǒng)計(jì),研究結(jié)果表明1不同品種的葉芽發(fā)芽情況差異很大,花葉丁香和羽葉丁香發(fā)芽較晚,比暴馬丁香發(fā)芽期晚將近一個(gè)月。其他幾個(gè)品種發(fā)芽去相差不多,幾乎都在三月下旬到四月上旬?;ㄈ~丁香和羽葉丁香的發(fā)芽期與展葉期相接較近,約一周左右,其余的品種發(fā)芽期與展葉期間隔較遠(yuǎn),約十五天左右。2園內(nèi)丁香開(kāi)花最早為紫丁香,約在5月1日左右,開(kāi)花最晚的為暴馬丁香,約6月16日左右,其花期可延續(xù)在7月9日左右,園內(nèi)丁香的群體花期可達(dá)70天左右。3對(duì)開(kāi)花期與花期接近的紫云丁香、羅蘭紫丁香、波峰丁香、什錦丁香、長(zhǎng)筒白丁香、香雪丁香、紫丁香7個(gè)品種的花期做了單因素方差分析,品種間差異顯著,香雪丁香、波峰丁香與什錦丁香差異不顯著,其花期最長(zhǎng)。羅蘭紫丁香與香雪丁香差異不顯著,羅蘭紫丁香、長(zhǎng)筒白丁香與紫丁香差異不顯著。紫云丁香、長(zhǎng)筒白丁香與紫丁香差異不顯著,其花期最短。4十四個(gè)品種中只有紫丁香、暴馬丁香、小葉丁香、遼東丁香和紅丁香正常結(jié)實(shí),而其他種類(lèi)不結(jié)實(shí)或偶爾結(jié)實(shí)。5丁香的落葉時(shí)間比較短,通常只有3到5天左右。丁香葉片全部能夠自然脫落,這也表明,它們能夠感知到光周期變化,對(duì)包頭地區(qū)的氣候狀況有較強(qiáng)的適應(yīng)能力。

簡(jiǎn)介:7761【3博士學(xué)位論文論文趔II型醫(yī)型坐E她鋤歪叫曲墊鹽建絲口2壁一鹽I塑堂生始F£盈塾筐一一F1‘女I韓寡|』1一J一一指1,救『『『J垃衛(wèi)當(dāng)盟控一學(xué)中IIIQH一~堋地J物翟一I巾牛L垸一二L趨11翌芒J逍一挺變I。IJ釘世五生互』JJ二』旦一浙江大學(xué)博士學(xué)位論文羅氏沼蝦兩種不同的熱休克蛋白70分子生物學(xué)特征和功能這種特殊的表達(dá)方式說(shuō)明MARHSP70在羅氏沼蝦熱損傷保護(hù)中起著極其重要的作用,同時(shí)我們推測(cè)MARHSP70這種獨(dú)特的功能可能與其N(xiāo)一末端和C末端的缺失結(jié)構(gòu)相關(guān)。為了進(jìn)一步分析這兩種分子在胞內(nèi)的表達(dá)情況,我們采用原位雜交和免疫組織化學(xué)在中樞神經(jīng)和肝胰腺兩種組織中進(jìn)行了表達(dá)定位研究,結(jié)果表明MARHSC70和熱誘導(dǎo)后的MARHSP70在絕大多數(shù)細(xì)胞都有表達(dá),尤其是在神經(jīng)分泌細(xì)胞和肝胰腺的R和F細(xì)胞中表達(dá)水平較高,并且熱誘導(dǎo)后的MARHSP70的表達(dá)水平遠(yuǎn)高于MARHSC70,這些結(jié)果進(jìn)一步表明MARHSP70在熱應(yīng)激保護(hù)中扮演著重要的角色,同時(shí)也說(shuō)明熱應(yīng)激條件下,絕大部分細(xì)胞快速啟動(dòng)這種基因的表達(dá)。為了進(jìn)一步探討羅氏沼蝦這種誘導(dǎo)型MAR_HSP70的功能,我們構(gòu)建了MARHSP70一PET一28A原核表達(dá)系統(tǒng),成功地表達(dá)了MARHSP70,發(fā)現(xiàn)在IPTG誘導(dǎo)5HR后,目的蛋白可達(dá)到細(xì)菌總蛋白的50%以上,同時(shí)通過(guò)免疫印跡技術(shù)證明了這種蛋白的免疫原性,而且通過(guò)優(yōu)化條件,采用低溫誘導(dǎo),獲得了一定量可溶性MARHSP70。在MARHSP70功能分析中,我們建立了MARHSP70的活性檢測(cè)系統(tǒng),在衛(wèi)COLIBL21DE3導(dǎo)入這一基因并誘導(dǎo)表達(dá),發(fā)現(xiàn)能夠提高熱刺激后細(xì)菌的生存率,這說(shuō)明M”一HSP70在一定程度上起到了對(duì)細(xì)菌的保護(hù)作用,這為誘導(dǎo)型HSP70S功能研究提供了很好的模型和思路??傊?,這是第一次從蝦類(lèi)中克隆到兩種熱休克蛋白70S基因,并研究了它們的表達(dá)特征及可能的功能,這兩個(gè)基因具有大多數(shù)物種HSP70S的共性,而更重要的是,它們自身的特殊性,這些特殊性對(duì)于認(rèn)識(shí)水生無(wú)脊椎動(dòng)物HSP70S的結(jié)構(gòu)及功能有著重要的理論價(jià)值和研究前景。關(guān)鍵詞羅氏沼蝦,熱休克蛋白70KDA,熱休克,表達(dá)分析和定位,原核表達(dá),活性檢測(cè)

簡(jiǎn)介:陜西師范大學(xué)碩士學(xué)位論文煙草超聲生物學(xué)效應(yīng)的研究姓名鄭媛申請(qǐng)學(xué)位級(jí)別碩士專(zhuān)業(yè)植物學(xué)指導(dǎo)教師肖婭萍20050501進(jìn)細(xì)胞生長(zhǎng),而大劑量的處理則會(huì)造成不可恢復(fù)的影響超聲誘導(dǎo)3MIN的細(xì)胞,超聲后立即計(jì)數(shù),其死亡率為67%,而超聲處理5MIN的細(xì)胞,處理后立即計(jì)數(shù)其死亡率卻為44%,處理7MIN的細(xì)胞。處理剛結(jié)束時(shí)的死亡率為72%;短時(shí)間的超聲處理3MIN可以促進(jìn)愈傷組織細(xì)胞的生長(zhǎng),而長(zhǎng)時(shí)間的處理誘導(dǎo)7RAIN,則使其生長(zhǎng)明顯受到抑制。5MIN超聲處理愈傷組織的褐化率最低,而3RAIN和7MIN超聲處理后愈傷組織的褐化率都較高,尤其以7RAIN處理愈傷組織褐化最嚴(yán)重3種劑量的超聲都能使一定比例的細(xì)胞發(fā)生凋亡,效果相同。5適宜的低強(qiáng)度超聲波作用于植物細(xì)胞時(shí),提高了細(xì)胞膜和細(xì)胞壁的穿透性,高強(qiáng)度超聲在液體媒質(zhì)中產(chǎn)生激烈而短暫的瞬態(tài)空化效應(yīng)。空化泡崩潰的瞬間,在空化泡周?chē)a(chǎn)生局部高溫高壓并伴有強(qiáng)烈的沖擊波,從而產(chǎn)生一系列物理化學(xué)效應(yīng)1從細(xì)胞的表面結(jié)構(gòu)來(lái)看,則7RAIN處理對(duì)細(xì)胞結(jié)構(gòu)損傷最大,產(chǎn)生許多空化泡狀結(jié)構(gòu)。細(xì)胞團(tuán)周邊細(xì)胞的表面變得粗糙,出現(xiàn)許多不規(guī)則顆粒突起,細(xì)胞核質(zhì)相混、胞質(zhì)破裂,甚至固縮,并出現(xiàn)大吞噬泡,細(xì)胞壁嚴(yán)重扭曲,質(zhì)壁分離更加嚴(yán)重,有的細(xì)胞幾乎不含內(nèi)含物。2經(jīng)超聲處理3MIN后的細(xì)胞結(jié)構(gòu)模糊,液泡化程度增加,有的細(xì)胞核和細(xì)胞質(zhì)被大液泡擠到細(xì)胞邊緣,細(xì)胞器更少。細(xì)胞壁扭曲、變形,發(fā)生部分質(zhì)壁分離。煙草懸浮細(xì)胞經(jīng)超聲處理7MIN后,電鏡觀察表明,細(xì)胞質(zhì)內(nèi)出現(xiàn)新的小體,細(xì)胞內(nèi)含物減少,液泡化程度增加;細(xì)胞壁嚴(yán)重扭曲,質(zhì)壁分離更加嚴(yán)重,細(xì)胞核產(chǎn)生不規(guī)則微核。關(guān)鍵詞煙草;組織培養(yǎng);超微結(jié)構(gòu);超聲N

簡(jiǎn)介:獨(dú)創(chuàng)性聲明本人聲明所呈交的論文是我個(gè)人在導(dǎo)師指導(dǎo)下進(jìn)行的研究工作及取得的研究成果。盡我所知,除了文中特別加以標(biāo)注和致謝的地方外,論文中不包含其他人已經(jīng)發(fā)表或撰寫(xiě)過(guò)的研究成果,也不包含為獲得寧夏大學(xué)或其它教育機(jī)構(gòu)的學(xué)位或證書(shū)而使用過(guò)的材料。與我一同工作的同志對(duì)本研究所做的任何貢獻(xiàn)均已在論文中作了明確的說(shuō)明并表示了謝意。研究生櫞至乳時(shí)間五,卜/年Y只兩日/關(guān)于論文使用授權(quán)的說(shuō)明本人完全了解寧夏大學(xué)有關(guān)保留、使用學(xué)位論文的規(guī)定,即學(xué)校有權(quán)保留送交論文的復(fù)印件和磁盤(pán),允許論文被查閱和借閱,可以采用影印、縮印或掃描等復(fù)制手段保存、匯編學(xué)位論文。同意寧夏大學(xué)可以用不同方式在不同媒體上發(fā)表、傳播學(xué)位論文的全部或部分內(nèi)容。保密的學(xué)位論文在解密后應(yīng)遵守此協(xié)議研究生簽名凄嘭眇導(dǎo)師簽名許夠時(shí)間力,.√帆二∥年歲乒’日F,一Q年J刪日/F摘要針對(duì)寧夏中部干旱區(qū)砂田西瓜廣泛種植,土地利用結(jié)構(gòu)改變導(dǎo)致的西瓜連作障礙廣泛存在的現(xiàn)象,設(shè)計(jì)了西瓜一甜瓜_西瓜M、西瓜_大豆÷西瓜S、西瓜_辣椒_西瓜P、西瓜_油葵_西瓜O4種輪作模式,研究四種輪作下土壤化學(xué)性狀、土壤微生物學(xué)性狀及土壤微生物功能多樣性等指標(biāo)的變化規(guī)律,探討了輪作模式調(diào)控連作障礙的可能作用機(jī)制。主要研究結(jié)果如下1、持續(xù)砂田連作使得引起西瓜枯萎病發(fā)生的鐮刀菌數(shù)量和枯萎病病情指數(shù)呈上升趨勢(shì),土壤PH也隨著連作年限的增逐年升高,而土壤肥力總體呈現(xiàn)下降趨勢(shì),西瓜產(chǎn)量在連作栽培條件下持續(xù)下降。2、持續(xù)砂田連作條件下西瓜根際土壤微生物總生物量、細(xì)菌數(shù)量、放線菌數(shù)量和細(xì)菌/真菌的值呈下降趨勢(shì),而真菌數(shù)量逐年上升;土壤微生物生物量碳、氮和土壤微生物呼吸強(qiáng)度隨著連作年限的增加逐年下降持續(xù)連作改變了土壤酶的活性,隨著連作年限的增加土壤脲酶和蔗糖酶逐年下降,土壤過(guò)氧化氫酶先上升后下降,土壤脲酶先下降后上升。3、輪作模式下,西瓜根際土壤PH均低于連作,土壤各養(yǎng)分較連作有不同程度的提高,其中M、S、P土壤有機(jī)質(zhì)、全氮、堿解氮、全磷、速效磷在輪作年際間均顯著增加。4、輪作模式下,西瓜根際土壤微生物總生物量、細(xì)菌數(shù)量和放線菌數(shù)量均高與連作,真菌數(shù)量顯著低于連作,輪作兩年間B/F的大小依次為PMSOCK;土壤酶活性各輪作模式較連作均有所提高,其中P增加顯著;M、S、P顯著提高了西瓜根際微生物碳、氮、微生物熵和土壤基礎(chǔ)呼吸強(qiáng)度。輪作模式P、S、M改變了西瓜根際微生物群落對(duì)主要碳源的利用強(qiáng)度,以P的增幅更為顯著,培養(yǎng)168H時(shí),P、S、M的AWCD值分別比連作升高了70.25%、2.19%、10.67%,輪作模式O卻降低了西瓜根際土壤微生物碳源轉(zhuǎn)化效率。砂田輪作后,經(jīng)過(guò)田間調(diào)查測(cè)產(chǎn)發(fā)現(xiàn),西瓜枯萎病發(fā)生的鐮刀菌數(shù)量和枯萎病病情指數(shù)較連作有所下降,西瓜產(chǎn)量量較連作增加。P、S、M輪作模式被認(rèn)為是有利于改善砂田西瓜連作栽培根際微生物環(huán)境,緩解西瓜連作障礙的栽培模式關(guān)鍵詞西瓜,連作,輪作,病害,平均顏色變化率AWCD

簡(jiǎn)介:中山大學(xué)博士學(xué)位論文R768138水稻第6染色體S5區(qū)基因克隆及分子生物學(xué)研究專(zhuān)業(yè)遺傳學(xué)博士生仲健導(dǎo)師王金發(fā)教授主席力膨≥委員釤7彩犯≠掀式釤澎吾五1皴力廣州中山大掌生命科掌掌院二00五年五月十八日仲健中山大學(xué)博士學(xué)位論文水稻第6染色體SS區(qū)基因克隆及分子生物學(xué)研究過(guò)RTPCR檢測(cè)發(fā)現(xiàn)這些轉(zhuǎn)基因株的OS6AP1基因的轉(zhuǎn)錄本在體內(nèi)的水平大大降低。RTPCR分析表明以硎尸』基因在野生型植株的多個(gè)組織中表達(dá),但在花粉中未能檢測(cè)到其表達(dá)。這些結(jié)果表明OS6AP1基因的功能是調(diào)節(jié)水稻根系的發(fā)育,并影響株高和開(kāi)花期。OSDUFL296基因由10個(gè)內(nèi)含子與11個(gè)外顯子組成,推算的編碼蛋白由854個(gè)氨基酸殘基組成,并預(yù)測(cè)在蛋白質(zhì)N一端含有一長(zhǎng)約60個(gè)氨基酸的名為DUFL296的保守結(jié)構(gòu)域,RTPCR分析表明該基因在所有檢測(cè)的組織中都有表達(dá),包括花、葉、莖、根、幼穗等,而且表達(dá)量較為均勻。通過(guò)RNA沉默,發(fā)現(xiàn)經(jīng)RNA沉默的轉(zhuǎn)基因水稻表現(xiàn)出較慢的生長(zhǎng)速率,目前轉(zhuǎn)化小苗表型變化仍在進(jìn)一步觀察中。OSDSP基因序列分析表明OSDSPL基因克隆全長(zhǎng)1119BP,推測(cè)的OSDSPL基因產(chǎn)物由225個(gè)氨基酸組成。EDNA序列與基因組序列分析表明該基因由5個(gè)外顯子及4個(gè)內(nèi)含子構(gòu)成,多個(gè)在線分析工具表明該蛋白質(zhì)屬于雙特異蛋白磷酸酶家族。我們將該基因序列提交NCBI數(shù)據(jù)庫(kù),得到接受并獲得序列號(hào)AY669072。關(guān)鍵詞水稻ORYZASATIVAL;廣親和品種WCVRNA沉默;OSGAP1GPI一錨定蛋白;LYSM結(jié)構(gòu)域;OSDSP;OSDUFL296
