1、劉倩：6 種曹蓿屬植物多倍體誘導及倍性鑒定研究6 種苜蓿屬植物多倍體誘導及倍性鑒定研究研究生：劉倩指導教師：魏臻武教授( 揚州大學動物科學與技術學院)摘要苜蓿屬( M e d i c a g o L i n n ．) 植物是廣泛分布于世界各地的優(yōu)良牧草來源，同時也是改良農(nóng)業(yè)生態(tài)的優(yōu)良植物。隨著近年來我國革產(chǎn)業(yè)的迅速崛起，對高產(chǎn)、優(yōu)質(zhì)、高抗的優(yōu)良苜蓿需求迫切。倍性研究是苜蓿屬植物育種工作的基礎，也是研究苜蓿屬植物遺傳特性的理論指導。本文以

2、苜蓿屬植物紫花苜蓿( 四季旺) ( M s a t i v aC V ．S i r i v e r ) 、蒺藜苜蓿( 脫t r u n c a t u l a ) 、金花菜( 毖p o l y m o r p h a ) 、天藍苜蓿( 磁l u p u l i n a ) 、多葉型苜蓿( Ms a t i v aC V ．M u l t i f o l i a r )以及江蘇省地方品種淮陰苜蓿( M s a t i v aC V ．H

3、u a i y i n ) 為材料，通過不同濃度秋水仙素對各苜蓿屬植物種子進行處理，建立秋水仙素誘變苜蓿屬植物多倍體技術體系，以獲得不同倍性水平苜蓿新種質(zhì)；并以四倍體紫花苜蓿為對照，對苜蓿加倍多倍體進行了植物學及細胞學特征觀察。旨在為培育高產(chǎn)、多抗、優(yōu)質(zhì)的苜蓿新品種提供理論依據(jù)，且對苜蓿屬種質(zhì)資源遺傳多樣性研究具有重要意義。主要研究結果如下：( 1 ) 秋水仙素處理萌動種子誘導后，萌發(fā)的幼芽發(fā)生明顯畸變：畸變幼芽的下胚軸發(fā)生扭曲，明顯增

4、粗成棒狀，且胚根生長點受到嚴重抑制，胚根尖端膨大呈球形，大部分幼苗生長停滯未能成苗而導致最終死亡；( 2 ) 單倍體苜蓿植株株高、分枝數(shù)、節(jié)間數(shù)、每枝葉片數(shù)均顯著低于四倍體對照植株( p < o ．0 5 ) ，葉面積約為四倍體植株的一半。田間篩選到的疑似八倍體苜蓿植株，其葉面積、株高、分枝數(shù)、每枝葉片數(shù)均顯著高于四倍體對照植株Q < o ．0 5 ) ，但節(jié)間數(shù)差異不顯著；( 3 ) 四倍體紫花苜蓿氣孔保衛(wèi)細胞葉綠體數(shù)較二

5、倍體紫花苜蓿增加5 6 ．0 2 ％，差異顯著( p < o ．0 5 ) ，而疑似八倍體紫花苜蓿氣孔保衛(wèi)細胞葉綠體數(shù)比四倍體植株增加了2 1 ．6 2 ％，差異顯著( p < 0 ．0 5 ) ；( 4 ) 用改進的常規(guī)壓片法對苜蓿屬植物進行根尖染色體計數(shù)，建立了適于苜蓿屬植物染色體制片的技術操作規(guī)程。經(jīng)染色體計數(shù)分析，供試的6 種苜蓿屬植物材料染色體數(shù)目分別為：紫花苜蓿( 四季旺) ( 2 n = 3 2 ) ，蒺藜苗蓿
