3、ssion Vector Construction of Potato NPR1 Gene Abstract Sunflower (Helianthus annuus L) is one of the most important oil crops in the world. Sclerotinia sclerotiorum is one of the most serious diseases in sunflower. Until
4、 now, the fully resistant material to Sclerotinia sclerotiorum has not been found in sunflower. Also the safe and effective measure to control Sclerotinia sclerotiorum is not available. Using inducer to induce resistance
5、 in plants to defend pathogens is an effective means. So β- aminobutyric acid (BABA), methyljasmonate(MeJA) and 1, 2, 3-benzothiadiazole - 7- carbothioic acid S- methyl ester (BTH) were used to treat sunflower plants. Th
6、en several important defense- associated enzyme activity were tested. We found that after the above three substances induction, the activities of phenylanine(PAL), polyphenol(PPO) , peroxidase(POD) and catalase(CAT) are
7、higher than those of untreated control. And after inoculation with Sclerotinia sclerotiorum, lesion areas of detached leaves treated with the above inducers were significantly smaller than control. This indicates that BA
8、BA, MeJA, and BTH can induce resistance to Sclerotinia sclerotiorum. Our research laid foundations for further study of induced resistant mechanisms and the application in sunflower production. Potato (Solanum tuberosum
9、L) is the fourth largest food crop in the world, its cultivation area and production is only after wheat, rice, and corn. Potato plants are susceptible to a wide variety of diseases. Systemic acquired resistance (SAR) is
10、 an indicible defense response that produced by local pathogen infection. It confers long- lasting protection against a broad spectrum of microorganisms and is thus a very promising tool to combat these potato diseases.
11、It is well- known that NPR1 (non- expresser of pathogenesis related genes 1) is the master regulator of salicylic acid- mediated SAR, worked as a co- transcriptional regulator. NPR1 can induces the expression of PR genes
12、 and produces resistance to pathogens. The NPR1 cDNA and its promoter segment were cloned from potato sequencing material. The binary vector was constructed and transformed into agrobacterium. This laid foundation for fu
