簡(jiǎn)介:山東農(nóng)業(yè)大學(xué)碩士學(xué)位論文科技文獻(xiàn)對(duì)園藝科技發(fā)展的作用研究姓名李燕申請(qǐng)學(xué)位級(jí)別碩士專業(yè)農(nóng)業(yè)推廣指導(dǎo)教師毛志泉20051210山東農(nóng)業(yè)大學(xué)碩士專業(yè)學(xué)位論史摘要在現(xiàn)代科學(xué)研究中,科研人員、試驗(yàn)手段、科技文獻(xiàn)是缺一不可的三大要素,而科技文獻(xiàn)的利用貫穿于科學(xué)研究整個(gè)過(guò)程,并發(fā)揮著重要的作用。目前,科技文獻(xiàn)對(duì)醫(yī)學(xué)、水利、森林培育等方面的發(fā)展作用的研究較多,但對(duì)園藝學(xué)的發(fā)展作用研究較少。為此,本研究調(diào)查了當(dāng)前科技文獻(xiàn)的分布、查閱情況及園藝學(xué)發(fā)展水平,目的是通過(guò)了解科技文獻(xiàn)的利用情況及園藝學(xué)科的發(fā)展?fàn)顩r,分析科技文獻(xiàn)對(duì)園藝科技發(fā)展的作用,更好地促進(jìn)園藝科技的發(fā)展。主要研究結(jié)果如下1從園藝學(xué)院的師生查閱類別來(lái)看,查閱農(nóng)業(yè)科學(xué)期刊次數(shù)最多;對(duì)利用率最高的農(nóng)業(yè)科學(xué)類期刊分布狀況進(jìn)一步統(tǒng)計(jì),發(fā)現(xiàn)農(nóng)業(yè)科學(xué)期刊利用類別主要分布在學(xué)報(bào)、綜合類農(nóng)業(yè)科學(xué)、農(nóng)業(yè)基礎(chǔ)科學(xué)等期刊上。2對(duì)科技文獻(xiàn)的利用情況進(jìn)行了調(diào)查研究,統(tǒng)計(jì)表明,隨著時(shí)代的發(fā)展,科研人員對(duì)科技文獻(xiàn)的利用在數(shù)量和質(zhì)量上都呈增長(zhǎng)趨勢(shì)園藝學(xué)院本科生主要是利用圖書期刊查閱文獻(xiàn),而研究生與科研人員主要是利用電子期刊。在電子期刊中,全文數(shù)據(jù)庫(kù)利用率最高。3對(duì)科技文獻(xiàn)與園藝科技的發(fā)展作用進(jìn)行了相關(guān)性分析。研究表明引文量、科技檔案的查閱量與論文的發(fā)表相關(guān)系數(shù)均達(dá)到了O96以上相關(guān)性較大,也就是說(shuō),科技文獻(xiàn)直接促進(jìn)了專著與論文的寫作;科研經(jīng)費(fèi)逐年增加,與引文量相關(guān)系數(shù)為098,相關(guān)性較大,進(jìn)而促進(jìn)了科研水平的提高;而引文量、科技檔案的查閱量與獲獎(jiǎng)成果數(shù)量相關(guān)系數(shù)為一090左右,相關(guān)性不大。4研究結(jié)果表明隨著科研工作的進(jìn)一步開展,科研人員對(duì)科技文獻(xiàn)的利用數(shù)量不斷增多,利用質(zhì)量也不斷提高,科技文獻(xiàn)對(duì)園藝科技整體水平的發(fā)展有明顯的促進(jìn)作用。5存在問(wèn)題目前,我國(guó)科研人員利用科技文獻(xiàn)的能力和狀況不容樂(lè)觀。主要表現(xiàn)為不善于全面利用各類文獻(xiàn)、不善于利用各種檢索工具

簡(jiǎn)介:上海交通大學(xué)博士學(xué)位論文園藝產(chǎn)品真空預(yù)冷過(guò)程溫度檢測(cè)與品質(zhì)變化研究姓名宋小勇申請(qǐng)學(xué)位級(jí)別博士專業(yè)制冷與低溫工程指導(dǎo)教師李云飛201103上海交通大學(xué)博士學(xué)位論文和PH值的影響最小。二、采用紅外熱成像技術(shù)對(duì)鮮切花真空預(yù)冷不同部位的溫度分布和變化進(jìn)行實(shí)時(shí)檢測(cè)。結(jié)果表明①月季切花不同部位的平均降溫速率不同內(nèi)層花瓣最慢、外層花瓣最快、而葉子和莖桿無(wú)明顯差異,內(nèi)層花瓣的預(yù)冷終點(diǎn)溫度是外層花瓣的31倍。非洲菊切花舌狀花瓣的平均降溫速率最快而莖桿最慢,莖桿的預(yù)冷終點(diǎn)溫度是舌狀花瓣的99倍。不論月季還是非洲菊切花,不同部位的冷卻速率大小順序在不同的預(yù)冷階段表現(xiàn)不一致,各個(gè)部位不同區(qū)域的溫度分布亦不均勻;②在相同預(yù)冷條件下,分別用紅外和熱電偶檢測(cè)月季和非洲菊切花溫度,紅外檢測(cè)溫度比熱電偶檢測(cè)溫度分別低206_450OC和455一104OC。說(shuō)明熱電偶檢測(cè)受固定方法、位置和水汽化的影響較大;③月季外層花瓣和非洲菊舌狀花瓣的外緣為易發(fā)生凍傷部位。這就要求在鮮切花真空預(yù)冷過(guò)程中密切監(jiān)測(cè)不同部位特別是花瓣外緣的溫度變化,在保證預(yù)冷效果的同時(shí)避免冷傷發(fā)生;④月季和非洲菊單支花在瓶插期間的鮮重、氣孔導(dǎo)度和蒸騰速率變化率均小于花束,平均預(yù)冷終點(diǎn)溫度低于花束,瓶插壽命長(zhǎng)于花束。延遲預(yù)冷對(duì)切花品質(zhì)影響顯著PO05。隨延遲時(shí)間增加,預(yù)冷終點(diǎn)溫度不斷升高,瓶插壽命顯著降低P005,花束主要品質(zhì)受延遲時(shí)間的影響程度大于單支花。在本實(shí)驗(yàn)條件下,室溫存放10H,兩種切花均失去市場(chǎng)價(jià)值。三、研究成膜型抗蒸騰劑對(duì)鮮切花真空預(yù)冷效果的影響。噴施抗蒸騰劑之后進(jìn)行瓶插實(shí)驗(yàn)可以降低切花失水、延緩花朵開放進(jìn)程、降低氣孔導(dǎo)度、減少丙二醛積累、維持細(xì)胞膜完整性,切花壽命比直接進(jìn)行瓶插實(shí)驗(yàn)延長(zhǎng)32%。抗蒸騰劑和真空預(yù)冷的聯(lián)合使用在快速去除田間熱的同時(shí)有效地解決了水分損失過(guò)多的問(wèn)題。其中,先噴施抗蒸騰劑再進(jìn)行真空預(yù)冷處理的月季切花,其瓶插實(shí)驗(yàn)效果明顯優(yōu)于真空預(yù)冷后再進(jìn)行抗蒸騰劑處理的切花P005,切花壽命比后者增加了12%。四、研究真空預(yù)冷對(duì)組織微觀結(jié)構(gòu)和物性的影響。①初步證實(shí)真空處理對(duì)洋蔥表皮微觀結(jié)構(gòu)造成了較大的影響②細(xì)胞膜系統(tǒng)在短時(shí)間內(nèi)承受著快速降壓和水分蒸發(fā)遷移微應(yīng)力,液泡和細(xì)胞膜損傷導(dǎo)致胞內(nèi)水分蒸發(fā)損失,從微觀層面解釋了組織水分過(guò)度損失的原因;③同時(shí)研究發(fā)現(xiàn),膜損傷造成了花色素苷和電解質(zhì)流失,導(dǎo)致細(xì)胞顏色減退以及電導(dǎo)率升高。質(zhì)膜和細(xì)胞壁之間的間隙隨真空室壓強(qiáng)降低呈現(xiàn)先減小后增大的趨勢(shì)④掃描電鏡觀察顯示細(xì)胞表面形態(tài)、細(xì)胞壁和胞間層發(fā)生了不同程度的形變,直接導(dǎo)致表皮組織抗張強(qiáng)度的降低;⑤改變降壓速率對(duì)洋蔥表皮細(xì)胞表面形態(tài)、質(zhì)膜和細(xì)胞壁間距、水分損失、相對(duì)電導(dǎo)率、顏色和機(jī)械性能有明顯影響PO05。在慢速和快速降壓條件下,微觀結(jié)構(gòu)和物理性質(zhì)發(fā)生了較大變化,而中速降壓處理引起的破壞作用很小。本文采用宏觀和微觀研究相結(jié)合,實(shí)時(shí)圖像分析與理化指標(biāo)檢測(cè)相結(jié)合的方法,分析比較紅外熱成像和傳統(tǒng)熱電偶溫度檢測(cè)結(jié)果及其對(duì)鮮切花后續(xù)貯藏特性的影響,第Ⅱ頁(yè)

簡(jiǎn)介:學(xué)校代碼10225學(xué)號(hào)S13146學(xué)位論文幾種園藝植物病毒檢測(cè)技術(shù)研究指導(dǎo)教師姓名申請(qǐng)學(xué)位級(jí)別論文提交日期授予學(xué)位單位付耕云楊洪一副教授東北林業(yè)大學(xué)碩士學(xué)科專業(yè)微生物學(xué)2013年4月論文答辯日期2013年6月東北林業(yè)大學(xué)授予學(xué)位日期2013年6月答辯委員會(huì)主席論文評(píng)閱人聾夕參妊素大學(xué)摘要摘要園藝植物是一類供人類食用或觀賞的植物。園藝植物病毒是嚴(yán)重影響園藝生產(chǎn)的重要因素之一。為了進(jìn)一步控制病毒危害,本研究旨在建立從園藝植物辣椒、蘋果、櫻桃中分離DSRNA或總RNA的方法,并應(yīng)用瓊脂糖凝膠電泳、RT.PCR技術(shù)、免疫捕獲PCR技術(shù)等方法對(duì)分離的核酸進(jìn)行鑒定,從而建立可有效檢測(cè)病毒的技術(shù)體系。此外,還分別針對(duì)不同試材如葉片、辣椒制品、種子中的病毒進(jìn)行了檢測(cè),并對(duì)辣椒病毒的滅活進(jìn)行了初步探索。1、對(duì)辣椒病毒DSRNA的提取進(jìn)行優(yōu)化,建立了成熟的提取辣椒病毒DSRNA的技術(shù)體系,通過(guò)瓊脂糖凝膠電泳獲得了特異的DSRNA片段。分別采用雙酶解方法和DNA凝膠回收試劑盒對(duì)DSRNA譜帶進(jìn)行純化,經(jīng)比較采用DNA凝膠回收試劑盒回收DSRNA譜帶比雙酶解法更好,可有效獲得單一的DSRNA片段。2、利用改進(jìn)的CTAB法提取辣椒制品中的總RNA,并建立RTPCR技術(shù)檢測(cè)辣椒輕斑駁病毒的技術(shù)體系。對(duì)辣椒制品辣椒醬、咸菜、辣椒粉中的辣椒輕斑駁病毒進(jìn)行檢測(cè),在一些辣椒制品中檢測(cè)到了辣椒輕斑駁病毒,說(shuō)明辣椒輕斑駁病毒較難以被破壞。將辣椒制品中的辣椒輕斑駁病毒經(jīng)摩擦接種轉(zhuǎn)移至煙草,可在煙草上顯示明顯病毒侵染癥狀,并進(jìn)一步經(jīng)RTPCR證實(shí)。從而說(shuō)明辣椒制品中含有的辣椒輕斑駁病毒可有效傳播及增殖。已有報(bào)道顯示辣椒輕斑駁病毒可能侵染人體并引起一系列的反應(yīng),因而食品中的辣椒輕斑駁病毒也需要被密切關(guān)注。3、利用RT.PCR技術(shù),對(duì)北京、沈陽(yáng)、哈爾濱三地的商品辣椒種子中的辣椒輕斑駁病毒的帶毒率進(jìn)行了分析,結(jié)果顯示40種種子中,共有15種種子被辣椒輕斑駁病毒感染,說(shuō)明當(dāng)前該病毒的危害及傳播已較嚴(yán)重。4、利用高溫干熱處理對(duì)辣椒種子內(nèi)的辣椒輕斑駁病毒進(jìn)行了滅活處理,結(jié)果顯示,70℃處理未見明顯滅活效果,75℃處理48H可滅活70%的病毒,且對(duì)種子的發(fā)芽率影響相對(duì)較小。該技術(shù)體系具有一定的應(yīng)用潛力。5、利用RT.PCR技術(shù)對(duì)蘋果病毒進(jìn)行了檢測(cè),建立了對(duì)3種蘋果病毒的檢測(cè)技術(shù)體系。對(duì)一些蘋果、毛櫻桃、杏樣本進(jìn)行了檢測(cè),結(jié)果顯示不同樣本中3種病毒侵染率皆較高,且存在較多混合侵染的現(xiàn)象。6、基于RT.PCR技術(shù)的假陰性及假陽(yáng)性、血清學(xué)技術(shù)的靈敏性差問(wèn)題,探索建立病毒的IC.RTPCR檢測(cè)技術(shù)體系。分別以辣椒輕斑駁病毒和蘋果莖痘病毒為試材,利用病毒特異抗血清包被薄壁管,捕捉感病葉片粗汁液中的病毒粒子,之后運(yùn)用RT.PCR方法對(duì)捕捉的病毒進(jìn)行特異性擴(kuò)增,最終建立了可穩(wěn)定檢測(cè)辣椒輕斑駁病毒和蘋果莖痘病毒的技術(shù)體系。關(guān)鍵詞DSRNA;RT.PCR;免疫捕捉;辣椒輕斑駁病毒;蘋果病毒

簡(jiǎn)介:ZHEJIANGAFUNIVERSITYDISSERTATIONFORTHEDEGREEOFMASTER,娥/THESTUDYONMETHODANDEFFECTOFEFFICIENTPIPELINECULTIVATINGOFHORTICULTUREPLANTSINURBANROOFENVIRONMENTCANDIDATEZHAOYINGIEIADVISERZHUZHUJUN,PROFESSORSPECIALTYLANDSCAPEPLANTSANDORNAMENTALHORICULTUREDATEOFSUBMISSIONJUNE25,2013ZHEJIANGAFUNIVERSITYLIN’AN,ZHEJIANGPROVINCE,PRCHINAJUNE,2013謄摘要摘要本文在國(guó)內(nèi)首次將管道式栽培設(shè)備應(yīng)用于城市屋頂環(huán)境,在不具備蔬菜土壤種植條件的城市建筑物平面屋頂上架設(shè)內(nèi)部填充有特種栽培基質(zhì)與滴灌設(shè)備的PVC栽培管道,種植如速生葉菜、羽衣甘藍(lán)、櫻桃番茄等兼顧食用、觀賞與經(jīng)濟(jì)效益的園藝作物,驗(yàn)證在城市建筑的閑置屋頂上將農(nóng)業(yè)生產(chǎn)與休閑觀光相結(jié)合的可行性。文章研究了與植物品種相對(duì)應(yīng)的栽培管道結(jié)構(gòu)與管網(wǎng)系統(tǒng)結(jié)構(gòu),栽培基質(zhì)理化性狀衰變的快速修正方式,循環(huán)高效利用自然資源進(jìn)行肥水精準(zhǔn)補(bǔ)給的各項(xiàng)參數(shù),以及其他能夠使作物快速生長(zhǎng)、穩(wěn)定產(chǎn)量的方法,為該種植模式的推廣提供理論基礎(chǔ)和應(yīng)用樣板。通過(guò)研究發(fā)現(xiàn),向單根栽培管中一次性釋放2005GCA州032、998GKN03、1343GKH2P04、494G尿素與985ML微量元素濃縮液,配合使用300MM植物生長(zhǎng)口間距,200MMCI徑栽培管,后期對(duì)管中基質(zhì)PH值與通氣孔隙度的修正,在僅使用收集的自然降水進(jìn)行灌溉與不進(jìn)行補(bǔ)肥的情況下,可于每年8月上旬至次年7月上旬連續(xù)種植一個(gè)批次“黃妃”櫻桃番茄與“紫鴿”羽衣甘藍(lán),兩個(gè)批次“早熟5號(hào)”速生小白菜等作物,使番茄、小白菜的單株產(chǎn)量、品質(zhì)及羽衣甘藍(lán)的觀賞效果及產(chǎn)量接近甚至超過(guò)在相同氣候環(huán)境下由傳統(tǒng)土壤種植的CK,達(dá)到兼顧觀賞與產(chǎn)生經(jīng)濟(jì)效益的目的。關(guān)鍵詞園藝植物,城市屋頂,管道栽培。
