簡(jiǎn)介:分類(lèi)號(hào)學(xué)校代碼10464UDC研究生學(xué)號(hào)201121503Z密級(jí)碩士專(zhuān)業(yè)學(xué)位論文碩士專(zhuān)業(yè)學(xué)位論文產(chǎn)纖維素酶菌株的分離、誘變及生物學(xué)特性研究學(xué)位申請(qǐng)人人董鶴娟指導(dǎo)教師師丁軻副教授合作教師師王明旻研究員專(zhuān)業(yè)學(xué)位類(lèi)專(zhuān)業(yè)學(xué)位類(lèi)別別獸醫(yī)碩士專(zhuān)業(yè)學(xué)位領(lǐng)域?qū)I(yè)學(xué)位領(lǐng)域2013年4月摘要論文題目論文題目產(chǎn)纖維素酶菌株的分離、誘變及生物特性研究產(chǎn)纖維素酶菌株的分離、誘變及生物特性研究專(zhuān)業(yè)業(yè)獸醫(yī)碩士獸醫(yī)碩士研究生生董鶴娟董鶴娟指導(dǎo)教師指導(dǎo)教師丁軻丁軻副教授副教授摘要本研究從河南省洛陽(yáng)、商丘、周口、漯河等8個(gè)地市周?chē)慕諚U、稻草垛下層510CM的土壤58份。在加有羧甲基纖維素鈉和微晶纖維素粉為底物的選擇性培養(yǎng)基上進(jìn)行產(chǎn)纖維素酶菌株的分離,利用剛果紅平板法篩選透明圈較大的菌株。采用3,5二硝基水楊酸法測(cè)定纖維素酶活力。對(duì)篩選出的菌株進(jìn)行菌落特征、顯微形態(tài)、生理生化試驗(yàn)和16SRDNA序列進(jìn)行鑒定確定其種屬。對(duì)篩選出的高產(chǎn)纖維素酶活的菌株進(jìn)行生物學(xué)特性的研究。使用紫外誘變以及亞硝酸鹽誘變對(duì)篩選的菌株進(jìn)行誘變,得到的菌株用3,5二硝基水楊酸法測(cè)定纖維素酶活力。將芽孢桿菌和酵母菌菌液按5的濃度接種于秸稈發(fā)酵培養(yǎng)基中,分別于72H、96H和120H取樣進(jìn)行測(cè)定。通過(guò)平板滴種法測(cè)定秸稈中微生物數(shù)量,測(cè)定降解后秸稈的粗纖維以及粗蛋白含量來(lái)衡量菌株的降解能力。試驗(yàn)結(jié)果表明從不同樣品中共分離出具有纖維素酶酶活的菌株42株,通過(guò)初篩篩選出10株酶活較高的菌株。測(cè)定濾紙酶活和羧甲基纖維素鈉酶活,得到一株二者活力都較高的菌株BLY02,分別為05351UML和03654UML。顯微鏡觀(guān)察表該菌株呈短桿狀,有芽孢形成,生理生化鑒定初步確定為芽孢桿菌屬的菌株,16SRDNA分子鑒定BLY02為地衣芽孢桿菌。對(duì)菌株BLY02進(jìn)行了生物學(xué)特性研究,發(fā)現(xiàn)最適其生長(zhǎng)溫度為37℃,最適生長(zhǎng)時(shí)間為24H,最適其生長(zhǎng)PH為50。將菌株BLY02作為出發(fā)菌株進(jìn)行紫外誘變和亞硝酸鹽誘變,酶活力分別為07187UML和07437UML,較出發(fā)菌株提高了3431和39。秸桿發(fā)酵后,在72H、96H、120H物料中的芽孢桿菌和酵母菌均較對(duì)照組差異極顯著(P005)。而物料中粗纖維的含量在120H時(shí)降解率最高可達(dá)1165%,粗纖維的含量比原料降低了39個(gè)百分點(diǎn);粗蛋白的含量在120H時(shí)提高率最高可達(dá)4480,粗蛋白的含量比后料提高了349個(gè)百分點(diǎn)。

簡(jiǎn)介:ANALYSISOFTHEBIOLOGICALTRAITSANDGENETICDIVERSITYOFTRIPLOIDOLIVEFLOUNDERPARALICHTHYSOLIVACEUSTHESISSUBMITTEDTO墓NGDAOAGRICULTURALUNIVERSITYINFULFILLMENTOFTHEREQUIREMENTFORTHEDEGREEOFMASTEROFAGRONOMY1一一LIUHUICOLLEGEOFANIMALSCIENCEANDVETERINARYMEDICINESUPERVISORLIZHENYOUFENGZHUXIANGPINGQINGDAOCHINAJUNE,2014青島農(nóng)業(yè)大學(xué)碩士學(xué)位論文摘要三倍體牙鲆生物學(xué)性狀和遺傳多樣性分析摘要本研究對(duì)冷休克誘導(dǎo)的三倍體及對(duì)照組牙鲆的生長(zhǎng)狀況、營(yíng)養(yǎng)成分以及骨骼形態(tài)進(jìn)行了比較和分析;同時(shí),采用微衛(wèi)星分子標(biāo)記技術(shù)對(duì)這兩個(gè)群體的遺傳多樣性進(jìn)行了研究。1、通過(guò)定期測(cè)量二倍體和三倍體的全長(zhǎng)、體重等生長(zhǎng)性狀,比較了不同倍性牙鲆的生長(zhǎng)情況。在孵化后的11個(gè)月內(nèi),三倍體的全長(zhǎng)和體重均高于二倍體,其中2、5、9月齡的三倍體全長(zhǎng)和體重均顯著高于二倍體尸005。2、用X光照相技術(shù)對(duì)牙鲆骨骼形態(tài)進(jìn)行觀(guān)察。發(fā)JEYY養(yǎng)殖條件下,二倍體對(duì)照組和三倍體組均有脊椎畸形現(xiàn)象,常見(jiàn)的脊椎畸形有4種椎體聚合、椎體單面壓縮、椎體雙面壓縮和椎體移位,其中椎體聚合是最常見(jiàn)的脊椎畸形,567%的三倍體和333%的二倍體都出現(xiàn)椎體聚合在二倍體和三倍體中,第816塊和尾部最后10塊區(qū)域內(nèi)的椎骨畸形率均較高??傮w來(lái)說(shuō),三倍體牙鲆的骨骼畸形率顯著高于二倍體三倍體,600%;二倍體,333%,P005,三倍體發(fā)生脊椎畸形的個(gè)體數(shù)和各種畸形出現(xiàn)的次數(shù)也均高于二倍體。3、對(duì)三倍體牙鲆肌肉的營(yíng)養(yǎng)成分進(jìn)行測(cè)定,并與二倍體進(jìn)行比較。結(jié)果表明,二倍體和三倍體肌肉營(yíng)養(yǎng)成分總體差異不大,其中水分、蛋白質(zhì)、脂肪、灰分含量均沒(méi)有顯著差異。二倍體和三倍體含有相同種類(lèi)的氨基酸和脂肪酸。三倍體肌肉中的蛋白質(zhì)、飽和脂肪酸、多不飽和脂肪酸和必需氨基酸含量高于二倍體,但是差異并不顯著。三倍體的1600、204∞6、EPA三種脂肪酸含量顯著低于二倍體儼005,THR、GLY、ARG、181C07、202國(guó)6、DHA含量顯著高于二倍體尸005。4、采用微衛(wèi)星遺傳標(biāo)記技術(shù)對(duì)冷休克誘導(dǎo)的三倍體群體及二倍體對(duì)照群體進(jìn)行比較分析,取培育至11月齡三倍體及同期二倍體對(duì)照矛鲆個(gè)體各32尾,通過(guò)高鹽法提取肌肉組織總DNA,利用篩選得到的21對(duì)微衛(wèi)星引物進(jìn)行PCR擴(kuò)增,并經(jīng)14%聚丙烯酰胺凝膠電泳進(jìn)行分型后,進(jìn)行人工判讀和分析。結(jié)果顯示,21個(gè)微衛(wèi)星位點(diǎn)均為多態(tài),二倍體和三倍體群體的平均等位基因數(shù)A、基因型6數(shù)、平均有效等位基因數(shù)池、平均觀(guān)測(cè)雜合度㈨和平均期望雜合度玩分別為56和50,190和142,37和28,0613和0868。0681和O629。二倍體和三倍體群體的多態(tài)信息含量PIC分別為O642和O589,個(gè)體識(shí)別率㈤為O660和0609,非父排除率㈣為0482和0399,其累積個(gè)體識(shí)別
