2、cious. The wool is white and excellent while the fringe is beautiful and the dermatome is light and soft. Combining the fur quality, this paper studies the molecular principle on lamb fur growing, which clarifies the cor
3、relation between the lamb fur character and the structural protein gene at the molecular level. This study not only has important significance on the theory, but also provides a scientific theoretical proof for nutrition
4、 regulating and breeding. This paper focuses on lamb fur of Tan Sheep for the first time, which analyzes polymorphism of the wool structural protein gene KAP1.3, selects the genotypes correlative to the fur quality and a
5、nalyzes cloning, sequencing, amino acid sequence and the secondary structure of the protein. The paper preliminarily discusses the molecular principle on which KAP1.3 gene regulates the character. 1. Gene KAP1.3 of Tan S
6、heep is of polymorphism. Furthermore, it has AA、AB and BB three genotypes and A、B two alleles. The frequency of allele A is higher than that of allele B. We reach a conclusion that Tan Sheep′s KAP1.3 gene locus accords w
7、ith Hardy-Weinberg equilibrium by χ2 test. He、PIC of the group are respectively 0.500 and 0.374, and the gene locus of Tan Sheep group are moderately polymorphic. 2. There is correlation between KAP1.3 gene of Tan Sheep
8、and the fur quality. The multiple comparison analysis shows that, in the wool curvature, the strand curve number of BB genotype is obviously larger than that of AA and AB genotype (P<0.05). The strand bending proporti
9、on of BB genotype is very obviously higher than AA genotype (P<0.01) and obviously higher than AB genotype (P<0.05); In the wool fiber length, the heterotypieal hair stretched length of BB genotype is obviously lon
10、ger than that of AA and AB genotype (P<0.05). The natural strand length and non-medullated wool stretched length of BB genotype are longer than that of AA and AB genotype, but there are not obvious differences among t
11、he three genotypes. In the proportion of the hair type, the hair number proportion of AA genotype is obvious different from that of AB genotype (P<0.05) while BB genotype is in-between. The hair type quality proportio
12、n of AA genotype is very different from that of AB genotype (P<0.01) while BB genotype is in-between. In wool fiber diameter, the diameter of heterotypieal hair of BB genotype is larger than AA and AB genotypes. The d
13、iameter of non-medullated wool is smaller than AA and AB genotypes. But the differences of the three genotypes are not obvious. The mutational KAP1.3 gene of Tan Sheep regulates the main fur quality by changing amino aci
14、d sequence and protein structure. Tan Sheep with KAP1.3 BB genotype change basic group T into C in coding region 194 bp, so that isoleucine(I) changes into threonine(T) in coding region 65aa. In the secondary structure o
15、f protein, the protein of AA genotype near 65 aa is β-folded while that of BB genotype is irregular folded. Transversion of the coding gene changes its corresponding amino acid sequence, which may be the hereditary basis
