簡介:西南醫(yī)科大學(xué)碩士學(xué)位論文中圖分類號中圖分類號R573;R7352;R4466碩士學(xué)位論文間皮素在胃癌中的表達(dá)及其生物學(xué)意義間皮素在胃癌中的表達(dá)及其生物學(xué)意義院(系)、所院(系)、所臨床醫(yī)學(xué)院研究生姓名研究生姓名王元學(xué)科、專學(xué)科、專業(yè)外科學(xué)導(dǎo)師姓導(dǎo)師姓名姜淮蕪學(xué)位類型學(xué)位類型科學(xué)學(xué)位二〇一七年二月〇一七年二月西南醫(yī)科大學(xué)碩士學(xué)位論文間皮素在胃癌中的表達(dá)及其生物學(xué)意義摘要背景及目的在消化系統(tǒng)腫瘤中,胃癌發(fā)病率高、死亡率高。世界衛(wèi)生組織的統(tǒng)計表明,胃癌的發(fā)病率在全球居于第5位。胃癌發(fā)病率在我國位列第2位,死亡率為第3位。我國胃癌患者確診時多已到中晚期。胃癌患者需要進(jìn)行綜合的治療已成基本共識,免疫治療、分子靶向治療在胃癌綜合治療中至關(guān)重要。間皮素是一種在細(xì)胞表面表達(dá)的糖蛋白,在腹膜腔、胸膜腔和心包腔的間皮細(xì)胞上間皮素有低表達(dá)。同時人們發(fā)現(xiàn)間皮素也在胰腺癌、惡性間皮瘤、卵巢癌等多種惡性腫瘤中過度表達(dá)。間皮素已在多個腫瘤靶向治療中成為熱點。據(jù)最新國外文獻(xiàn)報道,間皮素有望成為胃癌治療新靶點,而胃癌有顯著的異質(zhì)性,所以本實驗的主要目的是探索胃癌及淋巴結(jié)轉(zhuǎn)移灶間皮素的表達(dá)特點、間皮素的表達(dá)與臨床病理因素的關(guān)系、間皮素表達(dá)對患者生存狀況的影響,從而探討間皮素作為胃癌治療新靶點的價值、間皮素的生物學(xué)作用及間皮素對預(yù)后的預(yù)測作用。同時我們還會對自制組織芯片的可行性及價值進(jìn)行探索。方法采用回顧性研究的方法,隨機選取于2009年1月2011年1月在綿陽四0四醫(yī)院腹部外科已行胃癌根治術(shù)的80個病例納入實驗。從這些病例中取出80個腫瘤原發(fā)灶、相應(yīng)的55個轉(zhuǎn)移淋巴結(jié)和10個隨機陰性切緣的石蠟標(biāo)本;再隨機選取我院內(nèi)鏡中心的胃炎性病變石蠟標(biāo)本10個。將每個病例的性別、年齡、腫瘤大小、位置、病理類型、腫瘤浸潤深度、淋巴轉(zhuǎn)移、TNM分期錄入數(shù)據(jù)庫同時記錄患者5年隨訪資料的生存

簡介:目的與正常細(xì)胞比較,分析敲除IHH基因的軟骨細(xì)胞在體外培養(yǎng)過程中的力學(xué)生物學(xué)特性變化,明確IHH基因在軟骨細(xì)胞力學(xué)生物學(xué)特性維持作用。方法1、取剛出生的IHHFLFL純合并具有COL2A1CREERT2的基因工程小鼠40只,隨機分為實驗組與對照組,實驗組腹腔注射TM(TAMOXIFEN),對照組腹腔注射等量的植物油,連續(xù)注射5天后,急性分離肋軟骨細(xì)胞。2、實時熒光定量PCR(RTPCR)檢測IHH基因相對表達(dá)量,明確IHH基因的敲除效率。3、CCK8法和流式細(xì)胞術(shù)檢測同一時間點實驗組與對照組肋軟骨細(xì)胞的增殖和凋亡(N10)。4、RTPCR檢測COLI、GLI1、COLII、COLX、AGG和MMP13的MRNA表達(dá)水平的變化(N5)。5、微管吸吮技術(shù)及半無限體細(xì)胞力學(xué)模型測定IHH基因敲除后肋軟骨細(xì)胞力學(xué)特性的變化(N5)。結(jié)果1、CCK8檢測發(fā)現(xiàn),敲除IHH基因后,軟骨細(xì)胞增殖效率較對照組降低,其中48H、72H最為顯著,450NM波長吸光度分別為(0534±015)、(0722±013)(P2、流式細(xì)胞術(shù)檢分析發(fā)現(xiàn)第3天、第5天、第7天的實驗組細(xì)胞早期凋亡率較對照組均增加P3、RTPCR分析COLII和AGG的MRNA表達(dá)水平較對照組分別增高42倍(P4、微管吸吮及半無限體細(xì)胞力學(xué)模型測定軟骨細(xì)胞力學(xué)特性發(fā)現(xiàn)瞬時模量(E0)、平衡模量(E∞)、表觀黏性(Μ)均相對于對照組較高(P結(jié)論條件性敲除軟骨細(xì)胞IHH基因后能夠有效的提高軟骨細(xì)胞COLII和AGG的表達(dá),抑制COLX和MMP13的表達(dá)。并且軟骨細(xì)胞力學(xué)特性明顯的增加。提示,敲除INDIANHEDGEHOG基因?qū)τ趦?yōu)化種子細(xì)胞具有一定的價值。

簡介:目的脈絡(luò)叢上皮細(xì)胞除了分泌腦脊髓液,在中樞神經(jīng)系統(tǒng)中其它作用知之甚少,對綠色熒光裸小鼠脈絡(luò)叢上皮細(xì)胞進(jìn)行原代培養(yǎng),旨在開發(fā)出-種有自然熒光示蹤特征的工具細(xì)胞,并對其生物學(xué)特性進(jìn)行研究,為進(jìn)一步研究提供便利。方法解剖顯微鏡下提取裸小鼠脈絡(luò)叢組織,機械分離成細(xì)胞團(tuán),在盛有20%胎牛血清DMEM(F12培養(yǎng)基的多孔培養(yǎng)板中培養(yǎng),3天后第1次換液,之后每2天半量換液,1周后每3天換液1次,細(xì)胞貼壁后用CCK8及KI67測增殖活性,倒置熒光顯微鏡下觀察細(xì)胞形態(tài)和密度,TTR、S100,GFAP,ALPHASMA和NESTIN免疫細(xì)胞化學(xué)熒光染色對培養(yǎng)細(xì)胞類型進(jìn)行鑒定。結(jié)果培養(yǎng)的脈絡(luò)叢上皮細(xì)胞穩(wěn)定表達(dá)綠色熒光(EGFP),發(fā)出明亮的綠色熒光,在形態(tài)上呈多邊形,伸展寬廣,胞體大,幾乎每個細(xì)胞都高表達(dá)脈絡(luò)叢上皮細(xì)胞特異標(biāo)志TTR,但不表達(dá)GFAP,ALPHASMA和NESTIN,并且具有較強的增殖活性,在體外可連續(xù)傳代7代。結(jié)論本方法可成功培養(yǎng)表達(dá)TTR的綠色熒光裸小鼠脈絡(luò)叢上皮細(xì)胞,穩(wěn)定表達(dá)EGFP,有望作為具有自然示蹤特征的工具細(xì)胞用于相關(guān)實驗之中。
