3、CR(ReversetranionPCR).Mouserecombinantcalreticulin(rCRT)ispreparedbycloningCRTcDNAintopET15bvectexpressinginE.Coli.thenpurifiedbyNiNTAresin.MTTmethodisusedtotheappropriateconcentrationsactingtimesfapoptosisinducingdrugs.

4、DNAfragmentationelectropheticassayisusedtodetectcellapoptosisrealtimePCRWesternblottingassayareusedtodetectthechangeofCRTexpressionlevelinB16F1cellsafterdrugtreatment.Immunecellfluescencechemistry(ICC)isusedtodetectthesu

5、bcellulardistributionofCRTinB16F1cells.ThreedifferenttypesofapoptoticcellswereusedasimmunogeniccellstoinoculateBalbcmousetheyare:BENStreatedB16F1mitoxantronetreatedB16F1BENStreatedCRTpaintedB16F1.thentheinhibitioneffects

6、ofimmunoinoculationonthegrowthofB16F1livecellsinoculatedlatterlywereobserved.LDHreleaseassayisusedtodetectthekillrateofmousesplenocytesagainstliveB16F1cellsmouseH22cells.Results1)ThecDNAthatcontainsCRTFwasclonedsuccessfu

7、llyfromtheKMMouse.TherCRTwereexpressedinpurifiedfromE.coli.2)MTTassayshowedthatproliferationofB16F1cellswasinhibitedbyBENSmitoxantroneinatimeconcentrationdependentway.DNAfragmentationelectrophesisassayshowedthatbothBENSm

8、itoxantronehadabilitytoinduceapoptosisinB16F1cells.3)AccdingtotheresultsofRealtimePCRWesternblottingassaytheexpressionlevelofcalreticulingenewasnotchangedinB16F1cellsbyBENSmitoxantronetreatment.HoweverICCshowedthatmitoxa

9、ntronetreatmentresultedinthetranslocationofCRTfromcytoplasmtocellmembrane.NoobviousCRTtranslocationwasobservedinB16F1cellstreatedwithBENS.4)AnimalexperimentshowedthatinoculatingBalbcmousewithcellsthattreatedbymitoxantron
